No, it's not gone. But if it goes deep enough, it can be out of circulation for a while. The water in some aquifers is tens of thousands of years old, if not older.

The total amount of water on Earth is nearly constant. Plants use sunlight, water, and carbon dioxide to produce sugars and oxygen. Respiration (or combustion) consumes these sugars and oxygen to produce water, carbon dioxide, and energy.

A small amount of water is lost to space, while an even smaller amount of water arrives via space rocks.
